Formulation Of Pioglitazone Tablets With Some Innovative Techniques

This research work is aimed to formulate tablets with rapid release of pioglitazone. The drug pioglitazone is used in the treatment of diabetes. It belongs to class II as per biopharmaceutical classification system i.e., having low solubility and high permeability. The drug shows dissolution-rate limited absorption,having poor and variable bioavailability which needs enhancement of dissolution rate for maximum therapeutic efficacy. In order to improve the dissolution rate and thereby the absorption, orodispersible tablets of pioglitazone were prepared using different techniques. The new concepts such as microcapsules and compression coating which are normally designed for sustained/controlled/extended and enteric release are proposed for enhancing the dissolution rate of Pioglitazone. Similarly the taste masking polymer EudragitE100 is employed to improve the dissolution rate. The process of coacervation with change in pH of the liquid manufacturing vehicle is another novel concept introduced in this investigation. Comparison of different techniques were made and the results are appropriately discussed in the book.
